Effect of chlorogenic acid on intestinal inflammation,antioxidant status,and microbial community of young hens challenged with acute heat stress

Heat stress in poultry is deleterious to productive performance. Chlorogenic acid (CGA) exerts antibacterial, anti-inflammatory, and antioxidant properties. This study was conducted to evaluate the effects of dietary supplemental CGA on the intestinal health and cecal microbiota composition of young hens challenged with acute heat stress. 100-day-old Hy-line brown pullets were randomly divided into four groups. The control group (C) and heat stress group (HS) received a basal diet. HS + CGA₃₀₀ group and HS + CGA₆₀₀ group received a basal diet supplemented with 300- and 600-mg/kg CGA, respectively, for 2 weeks before heat stress exposure. Pullets of HS, HS + CGA₃₀₀, and HS + CGA₆₀₀ group were exposed to 38°C for 4 h while the control group was maintained at 25°C. In this study, dietary CGA supplementation had effect on mitigate the decreased T-AOC and T-SOD activities and the increasing of IL-1β and TNFα induced by acute heat stress. Dietary supplementation with 600 mg/kg CGA had better effect on increasing the relative abundance of beneficial bacterial genera, such as Rikenellaceae RC9_gut_group, Ruminococcaceae UCG-005, and Christensenellaceae R-7_group, and deceasing bacteria genera involved in inflammation, such as Sutterella species. Therefore, CGA can ameliorate acute heat stress damage through suppressing inflammation and improved antioxidant capacity and cecal microbiota composition.     

Effect of Jieyuwan on depression-like behaviors and expression of BDNF in hippocampus and prefrontal cortex of WKY rats

AIM:To investigate the mechanism of depression and its development, and to study the expression of brain-derived neurotrophic factor (BDNF) in the hippocampus and prefrontal cortex of Wistar-Kyoto (WKY) rats treated with Jieyuwan. METHODS:Adult male WKY rats were used as an animal model of endogenous depression. Wistar rats of the same strain were selected as control group. WKY rats were randomly divided into model group, citalopram group and Jieyuwan group. After intragastric administration for 21 d, the changes of depression-like behaviors were observed by sucrose preference test and forced swimming test. Immunofluorescence and Western blot were used to detect the expression of BDNF in the hippocampus and prefrontal cortex. RESULTS:WKY rats showed significant depression-like behaviors, and the expression of BDNF was significantly decreased in the hippocampus and prefrontal cortex (P<0.01). The reduction of neuronal axons in hippocampus was also observed. After drug treatment, the depression-like behaviors of WKY rats were significantly attenuated, and the expression of BDNF and the number of axons were increased (P<0.01). CONCLUSION:Jieyuwan effectively attenuates the depression-like behaviors of WKY rats, and BDNF is a key factor in its antidepressant effect. Our findings further confirm the involvement of BDNF in the development of depression.     

MD病毒抗独特型抗体的研制

制备鸡马立克氏病病毒(MDV)抗独特型抗体(Ab2),探讨Ab2模拟抗原诱导免疫应答的能力.用MDV免疫SPF鸡,分离提纯鸡抗MD-IgG(Ab1),Ab1免疫BALB/C鼠,用鼠脾细胞与SP2/0细胞制备Ab2杂交瘤细胞株,间接ELISA筛选阳性杂交瘤细胞株,经克隆、纯化2×株抗MDV独特型抗体杂交瘤细胞株(6E5,7A12),其中6E5培养上清、腹水ELISA效价分别为4000×、10000×,免疫20 d攻毒,具有抗MDV感染保护.     

猪细小病毒氢氧化铝疫苗研究

选用从国内浙江省某猪场流产死胎中分离的猪细小病毒强毒（简称浙PV－Z株），通过仔猪肾原代细胞增殖后，经甲醛灭活，再配以20％的氨氧化铝胶佐剂制成猪细小病毒氢氧化铝疫苗。该疫苗免疫性好，免疫程序简单。小剂量肌注豚鼠和猪，均能激起明显的抗体应答反应，疫苗对豚鼠接种2次HI价可达到1：320以上（通常1：640－1：1280）。对成年猪接种疫苗0．2ml,0.5ml,1.0ml免疫2次，或用2．0ml免疫1次，HI价范围在1：20－1：320之间，疫苗对胎儿的保护率可达到98．7％。实验室和田间跟踪试验显示该疫苗具有保存期长、耐热、安全、免疫保护率高等特点。

夏谷新品种豫谷6号的选育

豫谷６号是应用有性杂交技术，经连续南繁、温室加代选择育成的夏谷新品种。具有高产稳产、高抗倒伏、高度抗旱、综合抗病力强等优良特性，适宜中等以上肥力水、旱地种植。     

羔羊全营养代乳粉对羔羊增重影响的试验

为了研究羔羊出生后采取人工饲喂代乳粉溶液的效果，对10日龄羔羊称重，以平均日增重与母乳组比较，用t检验方法检验差异显著性。研究结果显示羔羊平均日增重试验组为0.097-0.126kg，母乳组为0.071-0.113kg，二者没有显著差异。在生产中，代乳粉能替代绵羊母乳饲喂羔羊，对多胎品系羊的推广应用以及解决绵羊缺乳、少乳或无乳问题具有重要意义。     

鸡白介素18基因原核表达质粒构建

根据已发表的江西土鸡白介素 -1 8(Ch IL-1 8) c DNA编码基因序列设计引物 ,用PCR技术从 p MDCh IL-1 8质粒扩增出编码鸡IL-1 8成熟蛋白基因 ,重组于 p BV2 2 0表达载体上 ,将重组质粒转化大肠杆菌 JM1 0 9(DE3 ) ,转化子经温度诱导的表达产物 ,SDS-PAGE电泳鉴定约 2万 ,N端开头 1 5个氨基酸序列测定分析 ,证明获得了鸡 IL-1 8成熟蛋白 ,为今后深入研究鸡 IL -1 8的生物学特性及其临床应用打下了基础     

三种不同方法显示仔猪小肠肌间神经元的比较

本研究采用小肠铺片NADPH-d组化法、NADH-d组化法和抗NF免疫组化法分别对0日龄、5日龄、28日龄仔猪的肌间神经元进行了比较研究。结果表明:NADPH-d组化法可以选择性的标记合成NO的神经元亚群,大部分阳性肌间神经元符合DogielI型神经元的形态特征;NADH-d组化法可以非选择性的标记大部分神经元,且采用该法观察到的肌间神经元的密度明显高于NADPH-d组化法,但该法不能清晰的显示肌间神经丛的细微结构,不适合用于神经元类型的观察;抗NF免疫组化法可以清晰的显示肌间神经元的突起,28日龄仔猪体积较大的阳性神经元主要为DogielI型和DogielII型神经元。
